PROTEINE ASSOCIEE A LA GESTATION OU PAG DE LAPINE. ISOLEMENT ET PURIFICATION PARTIELLE. MISE AU POINT D’UN DOSAGE RIA.

Résumé

Le placenta, zone de jonction du territoire maternel et fœtal, assure plusieurs fonctions:  de protection, de nutrition et de glande. Parmi les substances produites, se trouvent les protéines associées à la gestation ou PAG, qui constituent un marqueur précoce de la gestation et un indicateur du développement fœtal. Notre étude consiste à extraire ces protéines par précipitation au sulfate d’ammonium et à les isoler par chromatographie d’échange d’ions (DEAE-cellulose), à partir de placentas de lapines. Le dosage est réalisé par quatre systèmes radio-immunologiques (RIA) hétérologues, le standard et le traceur radioactif étant la PAG bovine. Les antisera utilisés sont l’antisérum anti PAG bovine, l’anti PAG ovine et deux anti PAG caprine. La protéine que nous avons extraite, apparentée aux PAGs, reconnaît les quatre antisera avec un meilleur résultat pour l’anti PAG ovine. Cette protéine placentaire, isolée et semi-purifiée, est retrouvée essentiellement dans la fraction 0,04 M NaCl en chromatographie anionique et ce, pour les quatre systèmes RIA utilisés.
