ETUDE IN VITRO D'UNE EXOPROTEASE D'ASPERGILLUS FUMIGATUS DEGRADANT LES PROTEINES CONSTITUTIVES DE MEMBRANES BASALES

Résumé

Aspergillus fumigatus est un champignon opportuniste responsable d'infections variées touchant principalement l'appareil respiratoire de l'homme et des animaux. Chez des sujets fortement immunodéprimés, l'aspergillose revêt souvent un caractère invasif, avec une évolution très rapide et généralement fatale. Depuis quelques années, des études considérables ont tenté d'élucider les mécanismes pathogéniques d'Aspergillus fumigatus qui président à son installation dans l'organisme hôte. En effet, il a été mis en évidence une corrélation entre la sécrétion m vitro par ce champignon d'une sérine-protéase de type chymotrypsine/subtilisine et ses capacités d'invasion tissulaire chez la souris. Nous avons étudié la dégradation de deux protéines constitutives des membranes basales, la laminine et le collagène type IV par cette enzyme. Notre étude a démontré au cours d'expériences réalisées in vitro la capacité de la protéase purifiée à dégrader les protéines constitutives de la membrane  basale.
