EFFET ANTI-LISTERIADE BIFIDOBACTERIUM INFANTISISOLÉ A PARTIR DE SELLES DE NOURRISSON ALLAITÉ AU SEIN

الملخص

Une souche de Bifidobacterium infantisa été isolée à partir de selles de nourrisson âgé de deux mois allaité exclusivement au sein de l’hôpital Ben Boulaid de Blida. Cette souche est douée d’une
activité antimicrobienne à l’égard de Listeria monocytogenes. Une purification partielle de la bactériocine a été effectuée d’abord par précipitation des protéines au sulfate d’ammonium puis par un passage du surnageant brut actif en chromatographie à échangeur de cations avec une élution de la bactériocine par paliers à 0,1, 0,3 et 1M en NaCl. À ce stade de purification l'activité antimicrobienne
a été observée dans la fraction éluée à 0,1M en NaCl. Puis par un passagede la fraction active précédente en chromatographie en phase inversée. La bactériocine a été soumise à l'action de
plusieurs enzymes protéolytiques (pepsine, trypsine, pronase et proteinase K). Ces dernières altèrent complètement l'activité antagoniste de la bactériocine. L'effet dutraitement thermique a montré que cette substance reste stable pendant 30 min de 70°C. jusqu'à 90°C. L'activité inhibitrice de la bactériocine reste stable dans des variations du pH acide et neutre. Par contre elle décroît dans des conditions de pH basique. L’effet anti-Listeriade Bifidobacterium infantisa montré après 6 jours de conservation que le taux de décroissance de la souche cible est de97,05 % à 15°C. Ce taux est plus important (99,50 %) lorsque l’espèce cible est conservée à 4°C.
